The B-cell antibody technology & Structural characterization group is responsible for the discovery of monoclonal antibodies that recognize infectious disease targets that are used to assess vaccine candidates. Flow cytometry of immune cells is an essential part in the generation and characterization of antibodies.

How You Will Participate

As a Senior Scientist, you will be embedded in our operations and your work will be part of the on-going vaccine programs to deliver first in-class medicines in the upcoming years. You will be an essential member of the team, focusing on flow cytometry-based applications (~50%) with the key task on sorting immune cells for discovery and manufacture of different types of antibodies. Experience in molecular biology for screening, cloning and production of monoclonal antibodies is preferred. Knowledge in protein purification, antibody characterization and liquid handling instruments would equalize lacking molecular biology experience. Routine participation in molecular cloning, cell culture maintenance and antibody generation/purification based on individual skill-set is expected.

Your current experience and your interest in learning existing and developing new methods for various applications including profiling of antibodies in blood circulation in combination with 10X genomics technology is essential. Your creativity and experience will be similar important than your ability to plan, coordinate and communicate with other teams in- and outside the organization. Your prior work experience, scientific judgement, willingness to integrate in existing processes and develop next level techniques is substantial for the team. With your scientific enthusiasm you will support the team in streamlining new technologies required to achieve new goals and to meet aggressive timelines to help our patients.

How You Will Achieve It
  • Execution of flow cytometry-based applications, with the focus on immune cell sorting for the generation of monoclonal antibodies by single B-cell cloning. Screening of clones using different fluorophores in a high-throughput manner. Contribution to new ideas and initiatives, and participation in project planning activities.
  • Execute identification and generation of monoclonal antibodies based on sequences from single B-cell cloning using various molecular biology methods such as restriction enzyme or Gibson/in-fusion/Golden Gate cloning. Screening of candidate antibody clones by ELISA or biolayer interferometry techniques is expected.
  • Supporting the team in antibody characterization using binding and functional assays after introduction into the methods.
  • Supporting integration of other technologies like yeast and/or phage display.
  • Present experimental results at internal group meetings and project meetings as required.
  • Maintain laboratory notebook in a detailed and descriptive manner. Keep current entries into the antibody and plasmid databases.
  • Perform daily, weekly and monthly laboratory activities, such as periodic monitoring of equipment, periodic laboratory cleaning and managing the laboratory waste
  • Satisfactorily complete all training in conformance with Departmental requirements. Where applicable, perform job responsibilities in compliance with cGMP/GLP and all other regulatory agency requirements.
  • Maintain a safe and productive work environment. Conducts all activities and makes decisions that are in accordance with Company policies & SOPs, Pfizer Values, & global regulatory guidelines, environmental guidelines, as appropriate.


BASIC QUALIFICATIONS
  • Master's Degree in Immunology, Molecular Biology or a related discipline and 7+ years of experience
  • Expertise working with microbial pathogens and sterile cell culture technique
  • Expertise in flow cytometry, especially cell sorting (FACS)
  • Proficiency in general molecular biology and immunology techniques
  • Familiarity with mammalian cell cultureas well asexpression systems for antibodies and recombinant proteins
  • Basic experience in cloning techniques, isolation of nucleic acids and construction/engineering of plasmid vectors
  • Experience in PCR reactions is expected.
  • Knowledge in antibody/protein purification techniques to compensate potential lack of molecular biology skills
  • Experience with Microsoft Office applications and relevant scientific software, including bioinformatics and graphing/statistical analysis tools is expected.
  • Ability to perform complex data analysis
  • Excellent oral and written communication skill


PREFERRED QUALIFICATIONS
  • PhD degree in Immunology, Molecular Biology or a related


Nice-to-Have
  • Hands on experience in the following areas is preferred:
  • Use of DNASTAR Lasergene/Genedata software,
  • ELISA or Luminex microplate assays,
  • Protein quantitation (UV/colorimetric) assays and SDS-PAGE/western blot analysis
  • EKTA based antibody purification (affinity, IEX, SEC)
  • Experience with automated liquid handler
