What You will Achieve

We are seeking a highly motivated individual with expertise in molecular biology and cell line generation to join the Cellular, Genomic, and Protein Sciences (CGPS) group. This individual will join a highly collaborative and cross-functional team of scientists that is responsible for the development of cellular models to drive discovery programs across diverse therapeutic areas. The core focus will be around d evelopment and characterization of physiologically relevant cell models. This will include working with primary cell models, development of clonal overexpression and gene edited cell lines, protein/gene expression characterization, and method and technology development.

How You Will Achieve It

The successful candidate will provide molecular and cell biology support for novel cell line generation and will execute standard cell culture processes including transfection, selection, sub-cloning, clonal expansion, and banking. The individual will regularly apply gene and protein expression characterization techniques to these lines and will support cell culture and provision, often at scale, to end users. He/she will also be involved in the production and support of more complex, disease relevant primary cellular models.

The successful candidates will be expected to:
  • Demonstrate basic laboratory skills to propagate and maintain various continuous and primary cell lines including preparation of medium and mycoplasma testing to ensure cell line sterility.
  • Be adept at handling human primary cell models and enable the growing application of more physiology relevant models for cellular assays.
  • Develop stable cell lines by conducting transfections, selection, single cell cloning, screening, characterization, and clone expansion.
  • Regularly apply techniques including DNA/RNA extraction, qRT-PCR, sequence analysis, FACS analysis, immunostaining, and sub-cellular fractionation/analysis.
  • Oversee the operation, maintenance, and troubleshooting of key technologies and instruments for cell line development and characterization.
  • Help to identify, evaluate, and, where applicable, implement novel methods or technologies that would enhance workflow efficiencies and/or improve the quality of cell reagent deliverables.
  • Analyze and interpret data, prepare cogent data summaries, and communicate results to team members.
  • Be highly motivated, highly organized and possess excellent verbal and written communication skills.
  • Show significant ability to work effectively in a matrixed, team-oriented environment.
  • Comply with all safety training and good laboratory practices.
  • Maintain up-to-date electronic laboratory notebooks and records in accordance with Pfizer policy and ensure proper archiving of banked cell lines.


Qualifications

Must Have
  • BS with at least 0-3 years relevant laboratory experience required
  • Highly proficient in aseptic mammalian cell culture
  • Experienced in the generation, characterization, and expansion of single-cell clones
  • Proficient in standard methods for cell characterization including mRNA and protein analysis such as qRT-PCR and Western blotting.
  • Excellent work planning, organization and record keeping is essential.


Nice to Have
  • Hands-on molecular biology experience including nucleic acid/protein isolation, genomic DNA analysis and sequencing.
  • Hands-on experience with kinetic imaging systems and/or immunostaining.
  • Previous experience with human primary cell models and/or iPSCs.
  • Demonstrated ability to effectively participate in cross functional collaboration and demonstrate excellent verbal and written communication skills.


PHYSICAL/MENTAL REQUIREMENTS
  • Sitting, standing, lifting, bending, and walking. Ability to perform practical laboratory bench work and work on a computer (while standing or sitting). Ability to perform mathematical calculations and interpret data.


NON-STANDARD WORK SCHEDULE, TRAVEL OR ENVIRONMENT REQUIREMENTS

  • Minimal, if any travel related to this position. Candidate may periodically be asked to travel to the Pfizer Cambridge, MA site to meet project team members.
  • Candidate must comply with laboratory safety practices and wear personal protection as required.
