What You Will Achieve

The purpose of this position is to implement achiral and chiral singleton and library auto-purification in support of small scale synthetic chemistry efforts in Groton. As part of the Automated Singletons for Automated Purification group (ASAP) in Discovery Sciences, this position involves adapting the purification workflow for enabling analytical and preparative scale HPLC/SFC with MS triggered detection to enable purification of final compounds for submission to biological testing. Close collaboration with other purification scientists will lead to process improvements that increase efficiency and scope of auto-purification adapted to the purification of compounds that are diverse in their structures, molecular weight, and physicochemical properties. The individual occupying this role will assist in problem solving and developing separation methods to enable purification of singletons and libraries, coupled with providing analytical data to confirm identity and purity, and developing a variety of sample handling methods to deliver the purified compound in a format suitable for further testing/evaluation.

How You Will Achieve It

  • Implement analytical and traditional scale high throughput singleton and library purification using the PureLab Autopurification platform and software.
  • Prequalify submitted samples for purification and develop methods for separation using analytical LC/MS or SFC/MS instrumentation.
  • Manage the purification workflow that includes robotics and liquid handling technology using the Autopurification workflow and software.
  • Characterize purified compounds using analytical HPLC/MS/ELSD or SFC/MS/ELSD techniques and submit samples for testing using the compound management infrastructure.
  • Collaborate with purification team members to optimize the purification workflow for efficiency and scope of samples that can be purified.


Qualifications

Must-Have

  • Bachelor's degree in chemistry (analytical or organic) with 2-5 years of industry experience or Master's degree in chemistry (analytical or organic) with 1-3 year industry experience required.
  • Experience using UPLC/MS instrumentation to characterize compound mixtures.
  • Experience using preparative HPLC purification for purifying small scale quantities of organic compounds.
  • Knowledge of Liquid Handling and Weighing systems is preferred.
  • Experience using high throughput library/singleton HPLC/SFC/MS purification methods.
  • Experience using Waters Masslynx software.


Nice-to-Have

  • Knowledge of Nano LC and quantitation techniques will be helpful.
  • Experience using UPC2/MS instrumentation to characterize compound mixtures.
  • Experience using preparative SFC purification for purifying small scale quantities of organic compounds.
