The Sr. Research Scientist will be a key member of the Discovery Group within Spark’s research organization and will participate in the advancement of Spark’s gene delivery platform. She/he will lead efforts to develop and characterize novel formulations and modifications of gene therapy vehicles.

He/she will be responsible for:

• Developing techniques and assays required for the analytical characterization of novel viral capsid and nanoparticle modifications.

• Characterizing gene therapy vehicles containing modifications including proteins, small molecules, nucleic acids, and peptides.

• Initiating and overseeing joint programs and collaborations with external parties.

• Writing study reports and manuscripts. Present results at appropriate scientific meetings.

Responsibilities

Job Function and Description

70% Develop techniques and assays required to establish analytical capabilities within the Discovery Group for comprehensive characterization of modified viral capsids and other gene delivery vehicles.

Characterize novel nanoparticle formulations and viral vectors, bioconjugates, and other chemical modifications to support pre-clinical studies. This includes executing experiments on the bench, extensive literature review, and championing product candidate development from proof of concept to IND submission.

15% Design experiments, organize, discuss, and record experimental data.

10% Generate technical reports, prepare presentation slides, generate novel concepts.

5% Trainings, lab meetings and administration work.

Education and Experience Requirements

• Typically has a PhD in Chemistry, Material Science, Engineering, or related field of study.

• Generally, has 3-5 years of experience related to biotherapeutics characterization.

• Familiarity with the generation and characterization of bioconjugates and nanoparticles.

• Experience with purification and characterization of nucleic acids is preferred.

• Experience with gene therapy and nucleic acid delivery is preferred.

Key Skills, Abilities, and Competencies

• Hands-on research experience with advanced biophysical and biochemical techniques required for biotherapeutic and nanoparticle analysis, such as:

• Chromatography (SEC, IEX, RP)

• Capillary electrophoresis (iCIEF, SDS-CGE)

• Biomolecular interaction (SPR, ITC)

• Spectroscopy (MS, UV/VIS, CD, intrinsic/extrinsic fluorescence, FTIR, Raman, NMR)

• Static and dynamic light scattering (SLS, DLS coupled to separations methods)

• Familiarity with various bioconjugation techniques, nanoparticle functionalization, and ligand-mediated targeting.

• Strong analytical skills and problem-solving capabilities.

• Proven ability to lead projects, work independently in hands-on laboratory setting, self-motivated to learn and develop new techniques, perform multiple tasks simultaneously, keep accurate records, and comply with company policies.

• Excellent communication skills (both oral and written).

• Works with colleagues as a team player.

• Strong organizational skills.

• Experience with Electronic Lab Notebook (ELN) and Laboratory Information Management Systems (LIMS) a plus.
