Site Name: USA - Pennsylvania - Upper Providence Posted Date: Aug 11 2021 The Investigator will be part of a drug discovery research team in Novel Human Genetics Research Unit at GSK R&D. This team focuses on the design and execution of preclinical studies to develop novel medicines for the treatment of chronic neurodegenerative diseases such as Alzheimer's, Frontotemporal Dementia (FTD), and Amyotrophic Lateral Sclerosis (ALS). The successful candidate is one who is self- and science-driven, collaborative, and technically resourceful. The candidate must have a comprehensive understanding of Alzheimer's disease, and utilize this knowledge to establish iPSC based cellular models of neurodegeneration and neuroinflammation. Experience in molecular and cell biology as well as biochemical methods is expected. The candidate will contribute to the development of preclinical models and utilize them in target ID/validation as well as profile molecules in neuropharmacology studies for drug discovery programs. Key Accountabilities: Establish relevant mechanistic in vitro iPSC derived cellular models to support preclinical drug discovery Conduct molecular (RNA isolation, qPCR, gene editing), biochemical (ELISA, MSD, Quanterix, Western) as well as iPSC derived cellular (FACS, live imaging) assessments. Define, design, and perform pharmacologic and/or genetic modification experiments in in vitro cellular models to validate targets and/or progress lead optimization projects for drug discovery Conduct pharmacological studies with an array of techniques including, but not limited to, stereotactic brain surgery, surgical methods to induce neurodegeneration and inflammation in the brain and spinal cord Independently execute laboratory experiments, conduct statistically supported data analysis, and deliver results in a timely manner to enable informed decision making Clearly record and communicate study results at internal and external meetings Commit to continual improvement by reading and applying the latest scientific literature, methodologies and technologies Comply with all standards and practices of data integrity, ethics, and safety Model GSK's values of patient-focus, respect, transparency, and integrity Why you? Basic Qualifications: We are looking for professionals with these required skills to achieve our goals: PhD or MD/PhD with 1-2 years' Post-Doctoral research experience in neuropharmacology or neuroscience in Alzheimer's Disease Demonstrated experience in gene editing, developing triculture model using iPSC derived neuronal and non-neuronal cells; versatile in cell biological methods such as FACS, imaging and basic biochemical and molecule biology methods. An integrated knowledge of neurodegenerative diseases, and prior working experience with neurodegenerative or neuroinflammatory diseases (e.g., Alzheimer's disease) Experience with statistical data analysis Preferred Qualifications: If you have the following characteristics, it would be a plus: 3+ years pharmaceutical or biotechnology industry research experience working in matrixed drug discovery project teams Knowledge and work experience in FTD and ALS in addition to Alzheimer's Disease Demonstrated experience of learning agility (conceptual, technical, team work) Outstanding written and oral communication skills including a strong record of scientific publications Interest and agility (technical, conceptual) to navigate into different disease areas beyond neurodegeneration (e.g. metabolic disease) About NHG RU: The Novel Human Genetics Research Unit (NHG RU) was formed in 2019 to identify and advance novel targets with genetic association to disease on the foundational observation that targets with human genetic evidence supporting their causal role in the intended indication improves clinical success by two-fold. The NHG RU is therapeutically broad and complements other Research Units with strategic focus in immunology and oncology. The current portfolio interest includes projects focused on neurological diseases, including Alzheimer's, Parkinson's, ALS, FTD, and Pain, in addition to metabolic diseases. The mandate of NHG RU is to identify novel targets based on human genetics, develop assets for clinical studies, and perform Phase 1 first time in human and proof of mechanism studies in human clinical studies. This goal is enabled through strategic external (23andMe, UK Biobank, FinnGen, Laboratory of Genomic Research, Public-Private Consortia) and GSK-internal partnerships (Functional Genomics, AI/ML, and Medicinal Science and Technologies Groups).
