BioMedicine Design's Endogenous Antibody Discovery Group is seeking a Scientist to expand and diversify lead panels of antibodies for biotherapeutic development. The candidate will be primarily responsible for carrying out microfluidics-based selections of B cells using the Berkely Lights Beacon platform. In addition, the candidate will work closely with colleagues to efficiently identify paired VH and VL sequences from selected hybridoma hits or single B cells using Sanger sequencing or multiplex NGS methods. The role also requires application of new technologies and workflows to support our ongoing commitment to enhance and streamline discovery of lead therapeutic antibody clones. The candidate will work in collaboration with BioMedicine Design's High Throughput Assay Screening group as well as colleagues in BioMedicine Design's Bioinformatics groups. The successful candidate will have a demonstrated track record in some of the following areas:
  • Antibody discovery experience with cell-based microfluidics selections.
  • Antibody sequence recovery from hybridoma or primary B cells with RT-PCR/vector construction
  • Antibody sequence recovery from hybridoma or primary B cells with Next Generation Sequencing.


ROLE RESPONSIBILITIES
  • Apply microfluidics-based methods to select antigen-specific B cells
  • Antibody sequence recovery from hybridoma or primary B cells
  • Isolation of primary cells from immunized rodents
  • Cell culture and transfection
  • Antibody specificity confirmation with ELISA and flow cytometry


Qualifications

Must-Have

  • Bachelor's Degree
  • 3+ years of experience
  • Substantial experience mammalian cell culture techniques
  • Strong expertise in molecular biology skills including vector construction, RT & PCR, NGS and extensive experience with sequence analysis using Vector NTI, Geneious or other softwares
  • Working knowledge of single B cell screening platforms, preferably with BLI Beacon platform
  • Ability to effectively interact and collaborate with other scientists and computational biologists, both within the group and in other groups
  • Excellent written and oral communication skills


Nice-to-Have

  • Master's degree and 3+ years of relevant experience
  • Hands-on experience with Berkeley Lights Beacon platform, preferably with immune cells.
  • Knowledge of mouse B cell biology and familiarity with B cell lineage analysis.
