ROLE SUMMARY

We are seeking a highly motivated individual with cellular and molecular biology expertise to join our Cellular, Genomics, and Protein Sciences group supporting early drug discovery efforts. The successful candidate will leverage their experience with a range of methods to evaluate the effects of small molecules and genetic perturbations on cellular endpoints, all towards the goal of identifying and advancing small molecule lead compounds for various drug discovery programs. The individual will become an integral member of project teams across a multiple of disease areas including Immunology, Internal Medicine, Rare Diseases, Oncology, and Vaccines.

ROLE RESPONSIBILITIES

The successful candidate will join a team of scientists within the Cellular, Genomic, and Protein Sciences group that is responsible for a range of functions including target identification and validation, as well as the establishment of cellular models and assays to help advance early discovery programs. This individual will apply a broad background in cell and molecular biology skills to support several early discovery programs from the target identification and validation stages through to hit-ID, and mechanistic studies.

Successful Candidates will be expected to:
  • Conduct pharmacology experiments to evaluate the impact of small molecule compounds on cellular endpoints.
  • Carry out cell culture to maintain and propagate cell lines as well as primary or stem cell-derived models.
  • Apply standard molecular biology methods to characterize cell models, and to study the effects of small molecules on cells. This ranges from evaluating mRNA and protein expression, to leveraging cell based assay readouts including reporter assays.
  • Utilize gene perturbation techniques such as CRISPR and RNAi methods to modify endogenous gene expression to study in impact on cell endpoints and to evaluate compound mechanisms of action.
  • Conduct mid- to high-throughput cell assays to screen compounds for desired effects on cell endpoints.
  • Characterize cell models by leveraging various methods to monitor marker gene and/or protein expression
  • Be a quick learner to flexibly support a range of portfolio projects with a broad set of cell and molecular tools.
  • Help to identify, evaluate, and, where applicable, implement novel methods or technologies that would enhance workflow efficiencies and/or improve the quality of cell reagent deliverables.
  • Analyze and interpret data, prepare cogent data summaries, and communicate results to partners in the Research Units and Partner Lines.
  • Maintain up-to-date electronic laboratory notebooks and records in accordance with Pfizer policy, and ensure proper archiving of banked cell vials.
  • Be highly motivated, organized and show significantly ability to work effectively in a team-oriented environment.

BASIC QUALIFICATIONS
  • BS or MS with 2-5 years relevant laboratory experience required
  • Hands-on experience working with cells, including aseptic technique, culture and propagation of a variety of cells, and experience with generating stable engineered cell lines.
  • Experience in the application of standard molecular biology methods including the isolation and quantification of DNA/RNA, western blotting, RT-PCR, ddPCR and standard PCR, primer and probe design.
  • Experience with gene perturbation methods including RNAi, CRISPR, and over-expression methods.
  • Experience with compound treatment of cells and cellular assays such as reporter-based assays or imaging assays.
  • Experience in the generation, characterization, and expansion of single-cell clones
  • Proficient in standard methods for cell characterization including mRNA and protein analysis including qRT-PCR, Western blotting, immunostaining, and FACs.


PREFERRED QUALIFICATIONS
  • Previous hands-on experience with cell automation equipment and/or instruments to conduct plate-based pharmacology assays.
  • Previous experience with assay formats such as HTRF/alphaLISA, HiBit, and other reporters to monitor cellular protein levels.
  • Prior experience evaluating compound treatment effects on cellular pharmacology
  • Excellent work planning, organization and record keeping is essential
  • Ability to multi-task and to balance bench and desk activities


PHYSICAL/MENTAL REQUIREMENTS

Sitting, standing, lifting, bending, and walking. Ability to perform practical laboratory bench work and work on a computer (while standing or sitting). Ability to perform mathematical calculations and interpret data.

NON-STANDARD WORK SCHEDULE, TRAVEL OR ENVIRONMENT REQUIREMENTS
  • Minimal, if any travel related to this position. Candidate may periodically be asked to travel to the Pfizer Cambridge, MA site to meet project team members.
