Replication Stress, Senescence and Aging

The National Institute on Aging (NIA), a major research component of the National Institutes of Health (NIH) and the Department of Health and Human Services (DHHS), is recruiting for a postdoctoral position in the Section on Gene Targeting, Laboratory of Molecular Gerontology.

Replication of the genome is hindered by numerous impediments, including alternate DNA structures, protein DNA complexes, and chemical adducts. Failure to overcome them can result in cell death, cellular senescence, and/or genomic rearrangements which may be cancer inducing. Understanding how normal and tumor cells respond to these challenges is the subject of intense investigation. Interstrand DNA crosslinks (ICLs) have been regarded as absolute blocks to replication, and crosslinking drugs are frequently used in cancer chemotherapy. However, we discovered a new pathway that enables cells to continue DNA synthesis without resolving the ICLs (Mol. Cell 52, 434, 2013; Cell Reports, 27, 1794, 2019). We are studying the mechanism of “replication traverse” of the ICL and the implications for genome stability, aging, and cancer therapy.

A postdoctoral position to participate in this program is available for an individual with training in chemistry, biochemistry, or molecular biology, and an interest in the cellular response to replication stress and genome integrity.

Candidates must have a Ph.D. or M.D. with less than five years of relevant postdoctoral experience. Salary is commensurate with experience and accomplishments.
