A 12-months research grant for activities to be carried out at Scuola Internazionale Superiore di Studi Avanzati (SISSA) with the aim of providing convincing proofs-of-principle that herpetoviral amplicon vector-mediated Emx2 gene delivery can effectively antagonize GBM in transplanted murine models, ideally leading to its full eradication. The Candidate will specifically contribute to achieve the following goals:
•defining a biosafe multiplicity of infection range, suitable to get robust herpetoviral infection of GBM cells in the absence of detectable damage to healthy neural tissue;
•quantifying the capability of GBM cells and healthy neural cells to host amplification of an EGFP-expressing herpetoviral amplicon vector assisted by oncolytic herpetoviruses;
•building an herpetoviral amplicon vector driving Emx2 overexpression;
•assessing the anti-GBM activity of such Emx2-epressing amplicon vector in vitro and in vivo.
