UNIVERSITY OF FLORENCE Analysis of the role of Notch in renal tumorigenesis
Dysregulation of the Notch pathway is a critical step in the generation of renal carcinomas independently from the histological subtype. To study renal tumorigenesis, we generated the Pax8.rtTA;TetO.Cre;R26.NICD1.GFP (Pax8/NICD1) mice, in which all renal tubular cells express the Notch1 intracellular domain. In these animals, we observed the generation of cystis and of hyperproliferative tubular structures. Aims of the project are: characterize the lesions through immunofluorescence, IHC and RNA analysis following laser microdissection of the lesions; identify the pathways modulated by Notch1 upregulation by trascriptome analysis of in vitro colture of renal tubular epithelial cells infected with the Notch1 intracellular domain.
