The research will aim at enriching marine dehalorespiring bacteria and studying the substrate specificity of their reductive dehalogenases. A marine microbial culture able to dehalogenate several organohalides, characterized by the presence of two dehalogenating species and several genes coding for reductive dehalogenases, will be used. The enrichment will be carried out using a number of carbon and energy sources, antibiotics and microbial inhibitors, and will be monitored via PCR-DGGE of 16S rDNA. The culture will be then grown on different organohalides and the reductive dehalogenase genes overexpressed in the presence of each substrate identified via RT-qPCR.
