In the field of “Extracellular Vesicles in Neural Cell Communication”, the research group of PD Dr Eva-Maria Krämer-Albers offers the following PhD project:

Glia to neuron exosome transfer in vivo

Although the delivery of exosomes from oligodendroctyes to neurons has been extensively studied in culture models, little is known about its prevalence in the brain in vivo. Exosome transfer can be traced in vivo by utilizing Cre-labelled exosomes and Cre-loxP mediated recombination of target cells. In this project, we use oligodendroglial CreERT2 driver mice to study spatio-temporal aspects of exosome transfer in the brain of reporter mice and to reveal the neuronal target populations and circuits involved. We will subject mice to sensory deprivation or stimuli such as enriched environment or motor activity to determine the contribution of neural activity. Furthermore, differential transcriptome analysis of exosome-receiving recombined neurons and non-recombined control neurons will be performed to assess exosome-dependent gene regulation in target neurons. Together, this study will reveal important insight into the prevalence of exosome-dependent neuron-glia communication in the brain and its functional relevance for neural homeostasis and plasticity. The project is integrated in the DFG Specific Priority Program 1757 "Functional Specializations of Glia" building an interdisciplinary network among the leading labs in Glia research in Germany.
