The genetic characterization of myeloid malignancies is underlying to investigate the molecular mechanisms involved in the pathogenesis, and has become critical for diagnosis, prognostic classification and treatment. Target therapy for a specific genetic lesion is already a reality for some types of leukemias. Instead, in other cases the heterogeneity and the complexity of the genetic abnormalities have not allowed the development or the consolidation of target therapy trials. The project will focus on molecular characterization of genetic alterations involved in the pathogenesis of myeloid malignancies. We purpose to study genetic rearrangements associated with poor prognosis, involving EVI1, PRDM16 and BCL11B genes. Furthermore, another aim will be the study of the mechanisms underlying the occurrence of monosomy 7. A deeper characterization of specific genetic changes should translate in the choice of best treatment and in the design of more targeted therapeutic trial.
