Much of Alzheimer's Disease (AD) research has investigated the amyloid cascade hypothesis, which postulates that AD is caused by changes in amyloid beta (Ab) stability and aggregation. Blocking Ab production by inhibiting the first protease required for its generation, beta-secretase/BACE-1, which is an aspartic protease, may be effective in blocking AD progression. The drawback of known BACE-1 inhibitors is their poor capability to cross the blood-brain barrier (BBB), limiting their potential in vivo. This project aims to design and synthesize novel heterocyclic compounds capable of inhibiting BACE-1 activity by selecting suitable functional groups for the interaction with key amino acids of the enzyme's active site.
