The research subject will include studies of proteins essential for redox regulations during glucose-stimulated insulin secretion (GSIS), determination of their contribution to the type 2 diabetic etiology when impaired. Molecular biology studies will be performed with insulinoma INS1E cells or beta cells sorted from the accutase-digested Langerhans islets. Diabetology studies will include glucose-tolerance tests, tests of various secretagogues or GSIS potentiation by GLP-1. Beta-cell-specific transcription factors will be also studied. The role of mitochondria-morphology-shaping proteins will be elucidated. Overall, we'll determine which proteins are essential for redox regulations during GSIS and relate new findings to the possible development of glucotoxicity- and lipotoxicity- related oxidative stress initiating type-2 diabetes.
