Department: Medicine

School: Medicine

Location: Wolstein Research Building – Main Campus

Supervisor Name and Title: Sanford Markowitz, MD, PhD, Professor

POSITION OBJECTIVE

The responsibilities of this position include planning and executing research projects within a laboratory focusing on cancer genetics. The individual will work closely with multi-investigator team consisting of clinicians and basic scientists within the Case GI Specialized Program of Research Excellence (SPORE) and the National Center for Regenerative Medicine (NCRM). The individual will lead a specific research team consisting of a graduate student and research assistant within the group.  This individual will focus on research involving gastrointestinal cancersand inflammatory bowel disease; specifically within the development of new therapeutics. This will involve continuing existing studies on the effects of using a specific inhibitor of 15-PGDH to improve recovery in murine colitis models.  Specific experimental studies will require the ability to conduct DSS colitis studies in mice, to isolate and fractionate different colonic cell populations, to analyze cytokine profiles in these cell populations, and to work with genetically engineered mice to determine the functional effects of cytokine mediators downstream of 15-PGDH.  Additional studies will involve parallel assays of tumorigenicity of these pathways involving AOM and AOM/DSS mouse models. The individual will be responsible for managing and designing individual experiments, reporting these in a consistent fashion and in a timely manner, managing issues of the scientific method including positive and negative controls, reproducibility, sample size estimate valuation, trends and troubleshooting, and refining techniques into standard operating procedures in the laboratory. Careful documentation and accountability is essential.  The individual will be responsible for writing and submitting manuscripts reporting research results.

ESSENTIAL FUNCTIONS

  1. Develop research projects involving characterizing the therapeutic benefit of specific inhibitors of 15-PGDH on murine colitis and the assessment of tumor induction or prevention. To further identify cytokine mediators of drug effect and the cell populations in which these mediators are expressed and then target.(30%)
  2. In vivo murine techniques including colitis and colon tumor models, cytokine assays, tissue fractionation and sample collection. In vitro techniques including flow cytometry, qRT-PCR, cell culture, Western blot, cell growth, in situ hybridization, etc. (25%)
  3. Manage research team consisting of graduate student and research assistant. (15%)
  4. Management of research collaboration between Case GI SPORE and National Center for Regenerative Medicine. (5%)
  5. Preparation and submission of publications of research and preparation and submission of mentored research awards in collaboration with the PI. (10%)
