Integrin ligands - anticancer drug conjugates

The PhD project will deal with the development of novel peptide drug

conjugates to target cancer stem cells from human tumors with a special

focus on colon and lung cancers. The ESR will first be involved in the

bioinformatic identification of peptide receptors in cancer stem cell

lines in collaboration with a bioinformatician. Several warheads will be

tested for their activity on cancer stem cell lines and peptide drug

conjugates or antibody drug conjugates targeting the identified

receptors will be generated and tested for their ability to affect cell

viability, growth and clonogenicity. The mechanism of action of the

compounds will be investigated with a special focus on subcellular

trafficking. If possible, mechanisms of drug resistance should also be

addressed using a large panel of cancer stem cell and commercial tumor

cell lines. The most promising agents will be tested in in vivo cancer

stem cell models (both orthotopic, subcutaneous and metastasis models).

The researcher who will take part in the above project will be

matriculated at the International Graduate School of Chemistry and

Biochemistry at Bielefeld University, Bielefeld, Germany.
